An Electron Microscopic Study on Mouse Liver after Acetaminophen Administration*

Abstract
The purpose of this study was to estimate the toxic effect of acetaminophen on hepatic cells of the mouse. The experiments were performed upon young male mice of 30g body weight. Acetaminophen. was administered by intragastric intubation as a freshly prepared suspension in distilled water. Mice received 125mg, 250mg, and 500 mg per kg body weight of acetaminophen, respectively, and were sacrificed 24 hours after administration of the drug.
The following results were obtained.
Light microscopy on liver tissue of the mouse received 125mg of the drug showed no detectable changes, but following the administration of the drug over 250mg revealed loss of glycogen, vacuolation, and necrosis in the hepatocytes mainly of centrilobular region.
Electron microscopy, generally, confirmed the light microscopic observations and revealed details of the changes of hepatocytes following administration of the drug.
Hepatocytes of the mice received 125mg of the drug exhibited glycogen areas with few glycogen particles and their negative images, slight increase of smooth endoplasmic reticulum, mitochondria containing the vesicles at the periphery, and rough¢¥ endoplasmic reticulum irregularly arranged and often vesiculated.
These fine structural changes of the hepatocytes were enhanced with 250ng of the drug, and furthermore fat droplets were significantly increased mainly in the glycogen areas.
Hepatocytes of the mice received 500nzg of the drug exhibited complete depletion of glycogen, aggregation of fat droplets, especially near and within the Golgi apparatus, and lysosomes with fragments of mitochondria, and showed necrotic changes in some cases.
